چکیده

Iodinated derivatives of adrenocorticotropic hormone (ACTH) were prepared with the use of NaI and chloramine-T and isolated by isoelectric focusing. The monoiodinated derivative (p1 7.6) was resolved into [monoiodoTyr2]ACTH and [monoiodoTyrZ3]ACTH by chromatography on columns of carboxymethylcellulose. Both monoiodo-AC’IH species exhibited only 30% of the maximum hormonal activity when assayed in an ACTH-sensitive adenylate cyclase system from adrenal tumor cells. The loss of hormonal activity was a consequence of oxidation rather than iodination. Iodination with lactoperoxidase instead of chloramine-T did not preserve hormonal activity. The monoiodinated derivatives of ACTH prepared with chloramine-T as oxidant were restored to full hormonal activity following reduction with cysteine in acetic acid. Mild acid hydrolysis accompanying the reduction reaction resulted in the conversion of the iodinated derivatives to products identified as [monoiodoTyr2]a1-24-ACTH and [monoiodoTyF]crl-24-AC’IH.

A Single-center Randomized Clinical Trial Comparing the Treatment Efficacy of High Dose Oral Prednisolone with Intramuscular Adrenocorticotropic Hormone in Patients with Infantile Spasm

Background Infantile spasm is a rare condition in infants from 4 to 7 months old. Treatment varies in different cases. Corticosteroids and adrenocorticotropic hormone (ACTH) are the most widely used treatment options; however, there are debates on their efficacy. The aim of our study is to compare corticosteroid treatment with ACTH in patients with infantile spasm. Materials and Me...

ACTH receptors in the adrenal: specific binding of ACTH-125I and its relation to adenyl cyclase.

Pure monoiodo ACTH-(125)I was prepared that was biologically active and free of unlabeled ACTH. Extracts of adrenal cortex that contained ACTH-sensitive adenyl cyclase, bound ACTH-(125)I; extracts that lacked the ACTH-sensitive cyclase did not bind ACTH-(125)I. Unlabeled ACTH inhibited the binding of ACTH-(125)I. Five ACTH derivatives which varied widely in biological activity were tested. All ...

Abnormal hormone responses of an adrenocortical cancer adenyl cyclase.

Properties of adenyl cyclase of normal adrenals and of a corticosterone-producing adrenal cancer of the rat have been compared. Enzyme activity was found in all particulate fractions of both tissues. The cyclase of the tumor as well as of the adrenals was stimulated by adrenocorticotropic hormone (ACTH) over similar concentration ranges. Unexpectedly, the tumor enzyme was also stimulated by epi...

Localization of an adrenocorticotropic hormone receptor on bovine adrenal cortical membranes.

The preparation of a subcellular fraction from bovine adrenal cortex is described. Characterization by electron microscopy, electron microscopic cytochemistry, and enzymatic assay indicates that this fraction is enriched plasma membranes. The preparation binds the steroidogenically active [Gin”] [W-Phe7] corticotropin, -ED amide. The specificity of this binding is demonstrated by competitive bi...

Hormonal regulation of focal adhesions in bovine adrenocortical cells: induction of paxillin dephosphorylation by adrenocorticotropic hormone.

A study of bovine adrenocortical cell shape on adrenocorticotropic hormone (ACTH) challenge showed that the cells round up and develop arborized processes. This effect was found to be (1) specific for ACTH because angiotensin II and basic fibroblast growth factor have no effect; (2) mediated by a cAMP-dependent pathway because forskolin reproduces the effect of the hormone; (3) inhibited by sod...
